FAQ

Q：可以连接大于 5kb 的片段吗？

A：大于 5kb 的没有测试过，可能比较难。

Q：反应过夜可以吗？

A：不可以，TOPO 异构酶具有限制性内切酶活性，时间过长会重新切割重组质粒，建议反应时间不超过 5min。

Q：Takara 的T 载体和我们TOPO 克隆试剂盒有什么区别？可以替代吗？

A：可以替代。区别：原理不一样，Takara 试剂盒利用的是 T4 连接酶，我们是 TOPO 异构酶，TOPO 异构酶更快更高效。

Q ：10909-Blunt Cloning Kit 和 10907- 零背景 TOPO-TA 克隆试剂盒中的 10 × Enhancer 是否一样吗？

A：是一样的，可以替换。

Q：自杀基因CCDB 和CCCTT 序列是什么？

A：1）自杀基因 CCDB 是指无片段插入该基因中时，该基因会表达蛋白杀死细菌，使克隆无法生长，CCDB 基因的序列是保密的。

2）CCCTT 序列是指拓扑异构酶能够偶联的序列。
