For CAR-T expansion with low IgG and exosome depletion, try to find domestic brands that specialize in cell therapy ancillary materials rather than focusing only on imported catalog numbers. First ask the supplier three things: the exosome depletion process (ultracentrifugation, precipitation, or tangential flow), whether the exosome particle count and CD63/CD81 are negative in the COA, and whether the IgG concentration is below your process requirements. If they have a local warehouse in Shanghai or ship from Suzhou/Hangzhou, it can usually arrive within a week. It is best to have them send a 100 ml trial size together with the QC report, then run one round of testing for expansion fold, viability, and CAR-positive rate before locking in the lot. Domestic exosome-depleted serum within 2500/500 ml is available, but lower-priced lots need more careful verification.

For your initial 200 ml and subsequent long-term use, it is recommended that you directly find a distributor in Shanghai with stock available. Require that the in-stock product must provide: a COA for exosome depletion and low IgG, endotoxin value, sterilization method by filtration, and lot number. Do not rely only on the product name; many so-called "low-exosome" products are not truly depleted, but are simply ordinary FBS centrifuged and the supernatant collected. Upon arrival, first perform a rapid test for residual exosomes, preferably using NTA or CD63 ELISA, and at the same time run a small-scale CAR-T comparison test. In procurement, you can negotiate safety stock and monthly shipments to avoid repeated freeze-thaw cycles. The three-week lead time for imported brands is indeed disruptive. If domestic products are in stock, first use 50-100 ml for validation, then decide on long-term replacement.
