Strictly speaking, if the cells are intended for reinfusion, the cryopreservation solution is classified as a formulation excipient and cannot be of ordinary research grade. Ordinary serum-free media may fail to meet reinfusion requirements in terms of sterility and endotoxin control. Clinical quality control personnel generally require GMP-grade or at least injection-grade or pharmacopeia-grade excipient sources. The difference lies not only in the composition but also in whether there is process validation, batch inspection reports, TSE/BSE statements, elemental impurities, residual solvents, and sterility or mycoplasma testing. If your budget is limited, you can first use GMP-grade material in small batches to lock down the process, and then discuss pricing. Keep the ordinary serum-free medium for process development and optimization as a control. It is not recommended to use ordinary serum-free medium to directly cryopreserve clinical reinfusion batches, as problems would be very difficult to explain.

In terms of composition, there may not be much difference, but ordinary serum-free media may use analytical-grade or research-grade raw materials, while GMP-grade generally requires pharmacopeial-grade or controlled raw materials, along with complete production records and sterility assurance. TSE/BSE statements are essential, especially if the formulation contains animal-derived components. Since your batch size of 100-200 ml is not large, you can choose small specifications of 100 ml or 50 ml to reduce the risk of repeated use after opening. Supply stability and documentation support are more important than price. It is recommended to ask several leading domestic manufacturers: whether they produce in GMP workshops, whether they can provide TSE/BSE statements, whether they have cases in CAR-T or immune cell cryopreservation, and whether they can provide clinical trial excipient support. Do not just take the sales representative's word for it.

GMP grade products with high cost-effectiveness are now a good choice for domestic production, don't just focus on imports. You can screen several conditions: having qualifications for cell therapy excipients or producing according to GMP standards; Provide complete COA, instruction manual, no animal source declaration, TSE/BSE declaration; Support small-sized spot goods. Because each batch contains 100-200ml, it is best not to buy large bottles and repeatedly pack them. We previously conducted a domestic GMP grade IIT and CAR-T cell cryopreservation using 100ml/bottle. After opening the bottle, it was packaged in one go, and the quality control teacher was able to accept it. Remember to ask the supplier to provide stability data and transportation cold chain solutions, especially in summer. Once clinical samples involve reinfusion, material risk is more critical than experimental data.

If CAR-T cells are used for reinfusion, it is not recommended to use ordinary scientific grade serum-free cryopreservation solution for clinical samples. Ordinary and GMP grade may have similar ingredients, but the difference lies in the quality system and documents: raw material grade, production workshop, sterile/endotoxin/mycoplasma testing, inter batch consistency, stability data, TSE/BSE declaration, change control, etc. IIT is indeed not as strict as drug registration, but ethics and quality control usually require key materials to be traceable. It is recommended to switch to GMP level or clearly label products that can be used for cell therapy production before entering clinical practice. Process development can start with regular, but clinical batches need to be changed, and bridging data should be retained.
