It is not convenient to name specific brands, but you can ask suppliers to provide reference users for primary neuron culture, as well as batch inspection reports showing endotoxin levels, and try to select batches with low values for hemoglobin, cortisol, estradiol, and total protein. Both major domestic manufacturers and imported brands offer low-endotoxin newborn calf serum, but whether it can be used for primary neurons still needs to be verified by yourself. Regarding lead time, domestic in-stock items are generally 3-5 days, while imported or custom batches may take 1-3 weeks. It is recommended to first apply for a 50 ml trial size, allow at least 2 weeks for preliminary experiments, do not wait until the day of formal isolation to open new serum, and confirm that the cold chain is intact.

If it is only for initial attachment with a later switch to serum-free medium, there is theoretically room to try, but the risk must be controlled. You can choose newborn bovine serum with low hemoglobin and low endotoxin and perform heat inactivation (56°C for 30 min) to reduce complement activity, while also lowering the serum concentration, for example, using 5% newborn bovine serum instead of 10% fetal bovine serum for attachment for 4-6 h, then switching to serum-free or B27-containing medium. However, primary neuron attachment also depends on poly-D-lysine or laminin coating, and serum is not the only variable. Same-batch controls must be run, recording the 2 h/24 h attachment rate, neurite outgrowth, and survival rate. If the attachment rate decreases by more than 15%, it is not recommended to continue.

Primary neurons are much more sensitive than tumor cells and hybridomas, so switching solely because the price is one-third is not recommended. The levels of endotoxin, hemoglobin, complement, and hormones in newborn calf serum are mostly higher than those in fetal bovine serum. Elevated hemoglobin may cause oxidative stress, and some primary neurons may show abnormal morphology after attachment, neurite retraction, or premature senescence. Differences in hormone levels may also affect spontaneous differentiation and the proportion of glial cells. If funding is tight, newborn calf serum can be used for prescreening and process exploration, but for key formal experiments, it is still recommended to retain fetal bovine serum and not proceed directly to the experimental group.
