When switching from CellTiter-Glo to a domestic alternative, do not replace everything all at once. It is recommended to use the same batch of cells and the same compound dilution plate, run the imported and domestic reagents in parallel, and compare the % inhibition rate or IC50 ranking for each plate rather than absolute RLU. Allow differences in absolute signal values, and focus on signal-to-noise ratio, inter-well CV, and the minimum distinguishable cell number. In addition, kinase inhibitor screening commonly uses DMSO, so the reagent's tolerance to 0.1%-0.5% DMSO needs to be tested, as fluctuations in DMSO concentration can affect luminescence readings. Remember to take into account differences in medium pH and phenol red color; phenol red may interfere, but the effect is generally small in luminescence methods, and you can ask the manufacturer whether they have data for compatibility with phenol red-free medium.

High throughput of 100 plates per week cannot be assessed by manual operation alone. It is recommended to validate on an automated liquid handling workstation: perform continuous dispensing into 96-well or 384-well plates and measure whole-plate CV. Due to the relatively high viscosity of the reagent, pay attention to residual reagent or bubbles in the dispensing tubing; the dispensed volume is adjustable. If domestic reagents contain different ratios of lysis/detection components, it is best to choose premixed single-component or A/B-component formats to reduce automation steps. The half-life must be sufficiently long. If your plate-reading sequence exceeds 20–30 minutes, choosing a product with a half-life greater than 2 hours can reduce edge effects. A 50 ml trial size is usually sufficient for 10–20 96-well plates. Use it first to evaluate well-to-well uniformity and the plate-reading time window, then decide whether to switch.

Inter-batch variation cannot be assessed based on the COA alone. It is recommended to request the manufacturer to provide samples or retention samples from three consecutive batches, retest them on the same cell plate, and calculate the CV of RLU and the signal-to-background ratio for each batch. If the inter-batch RLU difference is within two-fold and does not affect compound ranking, it is generally acceptable; if the absolute values fluctuate greatly, it is recommended to perform a cell number-signal pre-experiment calibration before using each batch of reagent. Bulk packaging is generally 500 ml or 1 L, with a lower unit price than small bottles. Based on your usage of 100 96-well plates per week, you can estimate the monthly consumption and then negotiate annual price locking. For lead time, most domestic products ship cold-chain within 1-3 days; imported aliquoted products are subject to availability, and note that the storage temperature must not involve repeated freeze-thaw cycles exceeding the number specified in the instructions.

For a domestic luminescence-based cell viability kit, first have the supplier provide a 50 ml trial sample, and use it to perform a gradient experiment: seed HEK293 and HepG2 at 500, 1,000, 2,000, 5,000, 10,000, and 20,000 cells/well respectively, with 6-8 replicate wells for each cell number, then after drug treatment lyse and read the plate according to the instructions. Check two things: whether low cell numbers are distinguishable above 3 times the SD of the blank, and whether high cell numbers have not exceeded the upper signal limit and plateaued. The kit's linear range should at least cover the cell density you actually screen; if it is close to the upper limit, the read time can be shortened or the sample volume reduced. To measure signal half-life: after adding the reagent, read the same plate at 0.5, 1, 2, and 3 hours, and record the RLU decay.
