For immunohistochemistry pens, commonly imported options include Vector ImmEdge, Dako Pen, or Abcam hydrophobic pens, which provide clear circle boundaries and resistance to buffers and DAB. Domestic pens are lower in price, but some have mediocre resistance to ethanol or xylene and can only be used after antigen retrieval and before color development. Do not use immunohistochemistry pens to draw circles before high-temperature retrieval, as they tend to spread at high temperatures and lose their water-repellent effect. It is recommended to first buy 1 imported pen and 2 domestic pens for comparison, drawing circles in different areas of the same slide, and after retrieval and color development steps, check whether the circles remain intact and whether they have spread.

In terms of price, imported anti-detachment slides are usually several times more expensive than domestic ones, while domestic positively charged slides mostly have a lower cost per slide; for specifics, you need to ask the regional distributor. There is a clear price gap between imported and domestic immunohistochemistry pens—one imported pen often costs as much as several domestic ones. A quantity of 200 slides and 5 pens is not large, so the room for negotiating a discount on its own is limited. It is advisable to purchase them together with antibodies and detection kits, or to ask a long-term reagent supplier for a discount under the category of commonly used laboratory consumables, which is more reliable than the retail prices from Taobao sellers.

For anti-detachment slides, don't just look at Taobao prices—focus on coating type and uniformity. Brain and adipose tissues tend to detach frequently, so prioritize positively charged slides or silanized slides. Common imported options include Superfrost Plus and Matsunami MAS-GP, which offer stable coatings but at higher prices. For domestic options, consider positively charged slides from major manufacturers such as Citotest and ZSGB-BIO, which are sufficient for routine IHC. After purchasing, don't rush into bulk use—first perform serial sectioning of the same tissue and test detachment rate and staining uniformity according to your high-pressure EDTA retrieval protocol before deciding whether to make a bulk purchase.

Tissue loss after retrieval is not always a slide problem. Section thickness, baking temperature, the degree of deparaffinization, and retrieval solution pH all affect adhesion. For brain tissue, I usually cut at 4 μm; for adipose tissue, 5–6 μm, and bake at 58–60°C for 2 hours to overnight. After retrieval, be sure to cool naturally; do not rinse with cold water to cool. If tissue loss still occurs, you can precoat slides yourself with poly-L-lysine or APES, but excessive coating thickness will increase background. Standardize the baking and retrieval procedures first, then change slides; otherwise, even imported slides may not solve the problem.
