We previously switched MSC primary cultures from South American to Australian serum. The first impression was that attachment and colony morphology were more stable, but proliferation was not noticeably improved every time; rather, batch-to-batch variation became smaller. Australian serum usually has stricter control of endotoxin and hemoglobin, and primary cells are relatively sensitive to these two parameters. Before switching, do not cut over completely all at once. First keep the existing South American lot for cryopreservation, then use the same Australian lot for parallel culture and check the 72 h attachment rate, doubling time, and surface markers. For 2 bottles of 500 mL, if the local distributor has stock, routine delivery is 3–7 working days; cross-region transfer takes 1–2 weeks; a single bottle can be negotiated at under 3000, but first-line original products may be stuck on tax and shipping. It is recommended to request a total price including tax and shipping and to keep a small sample.

From a budget perspective, staying within 3,000 per bottle is not impossible, but it depends on the brand and the definition of "Australian-origin." Some products labeled as Australian-origin are actually domestic aliquoting; you need to clarify origin quarantine and filtration records. Do not rely on verbal delivery timelines—ask the sales rep to confirm in writing the lot number, expiration date, and logistics. The main risk when switching MSC experimental serum is not the origin but the lot-to-lot variation, so it is best for the two bottles to be from the same lot. If the budget is tight, you can first take 1 bottle of Australian serum for trial use while keeping 1 bottle of the original South American lot as a fallback. For differentiation experiments, it is recommended to run the entire experiment with the same lot of serum to avoid interference caused by changing lots midway.

In our own experience, switching from South American to Australian origin has a positive effect on the long-term passaging and differentiation induction of primary MSCs, especially for adipogenic and osteogenic induction, where background differentiation tends to be more stable, but Australian origin is not mandatory. Focus on the COA: endotoxin, hemoglobin, total protein, pH, osmolality, and whether cell culture performance testing has been performed. If a supplier claims low endotoxin but cannot provide the original manufacturer's COA, be cautious. Regarding lead time, in-stock items from the northern warehouse are generally within one week, while original manufacturer orders may take 4–6 weeks. It is recommended to request the in-stock list first before making a decision.
