For non-original deep well plates, the main factors to consider are well shape, height, conical bottom, chemical resistance, and magnetic rod spacing. Most standard 96-well 2.2 mL deep well plates from domestic brands such as NEST and Axygen can fit, but some instrument slots have positioning notches designed for original plates, and even a slight height difference may cause the magnetic rod to press against the bottom or leave residual liquid after aspiration. It is recommended to first run an extraction protocol with empty plates from domestic brands, observe lid compression and mixing, and then compare 5-10 known samples in parallel with original plates, focusing on differences in nucleic acid concentration, A260/280, and downstream PCR results.

If reaction tubes are used for qPCR, the instrument-specified 8-tube strips are often matched to the heated-lid pressure and optical path. For non-original tubes, confirm wall thickness, light transmittance, and cap flatness; otherwise, fluorescence values will deviate. It is recommended to run a parallel comparison: same positive template, same reagent, original tubes versus non-original tubes, with 8–12 replicate wells per tube, and check the CV of Ct values and fluorescence values. Domestic compatible tubes are available from Axygen, NEST, and BioScience, but the specific catalog numbers must be checked against the instrument model using a compatibility table or confirmed in writing by the supplier; do not rely solely on the vendor's claim that they can substitute.

Do not substitute regular PCR film for sealing film. The extraction process involves heating and organic reagents, so the film must be resistant to DMSO or isopropanol, and its adhesive must withstand shaking and negative pressure. Poor sealing can cause evaporation or cross-contamination. You can choose silicone mats or aluminum films from NEST, Axygen, etc., but you must have the supplier provide samples to test the catalog numbers. When testing, measure evaporation: weigh before and after extraction or add dye to check for leakage. Original manufacturer products are indeed expensive, but once sample volume increases, you can negotiate tiered pricing with the manufacturer; it is not necessary to take risks just to save money on film.
