Incomplete Fc blocking is very common. Confirm that you are using CD16/32 monoclonal antibody, and make sure the concentration and incubation time are sufficient. I usually perform pre-blocking at 4 °C for 15 min; adding 1%–2% FBS or BSA to the staining buffer also helps. If splenocyte viability is poor or there is a lot of debris, nonspecific binding will increase markedly, so first ensure the viability of the single-cell suspension. You may additionally add 1%–2% normal mouse serum for blocking, but be careful not to affect subsequent surface marker staining.

F(ab')2 fragment secondary antibodies can significantly reduce non-specific binding via Fc receptors. Splenocytes are rich in Fc receptors, so they should be given priority consideration. However, even when using F(ab')2, it should still be combined with Fc block and cross-adsorption. F(ab')2 is usually a bit more expensive than intact IgG, and small flow cytometry pack sizes are relatively easier to find. During staining, antibodies should be titrated; do not use the manufacturer's recommended amount by default, as different splenocyte states will vary. Include a control with secondary antibody only, which can directly show whether F(ab')2 reduces background.

Control setup can help you determine the source of background. Prepare at least three tubes: unstained, secondary antibody only (PE), and isotype control primary antibody + PE secondary antibody. If the secondary antibody-only tube shows a high positive rate, it indicates direct binding of the secondary antibody or Fc binding; if the isotype control shows high background, switch to an isotype-matched control antibody. When nonspecific signal is high, do not blindly add compensation; first exclude dead cells by using a viability dye or 7-AAD to gate on live cells. Halve the secondary antibody concentration to observe changes in background, then perform titration.

When performing indirect flow cytometry with rat anti-mouse CD4, if the PE anti-rat secondary antibody has not been adsorbed against mouse IgG, it will directly recognize mouse Ig on the surface of splenic B cells, and high background is almost unavoidable. Therefore, you must choose a secondary antibody that has been validated by cross-adsorption against mouse IgG; product descriptions generally state "pre-adsorbed against mouse IgG" or similar. Before ordering, tell the sales representative the host species of the primary antibody and the rat Ig isotype for confirmation. This is not a nice-to-have, it is a must.

This type of PE anti-rat secondary antibody, 100 tests, imported, costs about 800 to 1,500 RMB; domestic options are lower, depending on the brand and distributor discount. When requesting a quote, confirm whether the definition of "test" is based on a 100 μL staining system or the manufacturer's recommended concentration. Many brands offer 25/50 test small packs; it is recommended to first obtain a small pack for titration and controls. Lead time: domestic in-stock is generally 1–3 days; imported replenishment takes 5–10 days. For trial sample applications, you can contact the distributor or the brand's technical support; there are usually promotions.
