稀释时用的水非常关键。10×PBS母液用于细胞间，建议用注射用水、细胞培养级超纯水或无热原水，不建议用普通纯水甚至自来水。普通纯水可能含有离子、有机污染物或热原，稀释后虽然过滤除菌，但不能有效去除全部内毒素，可能影响细胞状态。动物房也至少用纯化水或蒸馏水，避免使用未经处理的水。
