更稳妥的做法是把胶原酶粉末在干燥条件下按单次用量称量分装，-20或4℃干燥保存，临用前再用缓冲液溶解。这样比母液冻存更稳定，也不会为图省事损失活率。若确实要用母液，建议每次解冻后留少量做活性对照，与新配酶比较消化时间和得率。

保存缓冲液推荐含Ca²⁺/Mg²⁺的HBSS，加HEPES调pH 7.4；不要用含EDTA的PBS或普通PBS长时间保存。过滤除菌后再分装。10×母液比工作液更浓，冻存后有时会出现少量盐析，使用前轻轻混匀并观察，不建议过滤后直接再冻。

配好的胶原酶溶液我不建议长期冻存，尤其-20保存后容易出现活性下降和沉淀。我们实验室如果要冻，就用-80、单次用量分装，一般控制在1个月内用完，最长不超过3个月，并且用前取一管做小规模质控。反复冻融影响很大，同一管不要反复冻。

分装规格按一次小鼠胰岛消化量来，例如每次10×母液加0.1-0.2 mL到工作液，就分装成100-200 µL/管，避免顶空过大。用低吸附冻存管或螺旋管，管壁标批号、浓度、缓冲液和日期。每次取一管，不需要反复融。
