For liver tissue and phosphorylation detection, it is recommended not to focus only on the cocktail; the types and concentrations of phosphatase inhibitors in the formulation are more critical. In addition to the cocktail, I generally supplement sodium orthovanadate, β-glycerophosphate, and sodium fluoride to working concentrations, and use lysis buffer containing EDTA to simultaneously inhibit metalloproteases; if subsequent mass spectrometry is planned, switch to an EDTA-free version. Operationally, homogenize on ice, and immediately after lysis, centrifuge, quantify, and denature; do not leave samples at 4°C for too long. Regarding suppliers, for next-day delivery in Shanghai, you can check Bimake and MCE phosphatase inhibitors/cocktails, which usually are in stock, with 100× within 300. If conditions permit, ask sales to provide a small sample from the same lot to first test phosphorylation band stability, then place a bulk order.

Rather than just switching brands, I focus more on batch verification and storage. Repeated freeze-thaw cycles of the cocktail will significantly affect inhibitory activity. It is recommended to aliquot at 20-50 μL per tube upon arrival, store at -20 or -80, and take one tube each time. For a new batch, first lyse the same liver sample side by side with the old batch and run phosphorylation targets on the same membrane. Use it for samples only after the difference is acceptable. Choose suppliers that can provide a CoA or batch quality control information. Imported Roche, Sigma, and domestic Beyotime and Epizyme can all be purchased within budget. Roche may be slightly more expensive if in stock, and you can ask a local distributor. Liver samples contain more blood, so before lysis it is best to wash with cold PBS or perfuse in advance to reduce hemoglobin and protease release.

We recommend prioritizing brands with local warehouses in Shanghai or warehouses in East China. MCE, Beyotime, Epizyme, and Bimake are all common choices. When we extract protein from liver tissue, we use MCE's 100× cocktail together with separate phosphatase inhibitors/PMSF. Orders placed on weekdays from MCE's Shanghai warehouse generally arrive the next day, and a single 100× vial is within your budget. Your previous unstable phosphorylation may not necessarily be due only to batch-to-batch variation in the cocktail. Liver has high phosphatase activity, and the phosphatase inhibitor components in the cocktail may not be sufficient. We recommend adding an additional tube of phosphatase inhibitor cocktail. After arrival, use the same batch of liver tissue lysate to run a parallel comparison between the old and new batches, confirm that the difference in the target phosphorylated band/total protein ratio is not significant, and then process thirty to forty samples in bulk.
