Clinical serum matrix is complex. In addition to the dilution factor, it is recommended that you perform a spike recovery and linear dilution validation: dilute a known spiked serum sample 2-, 4-, and 8-fold and check the deviation between measured and theoretical values. If the instructions state "no dilution required," but your sample source is inflammatory patients, high values may hook, so a 2-fold dilution is instead protective. Include both high and low quality controls on each plate. Operationally, centrifuge serum samples after thawing to avoid fibrin clots. Do not use full-plate water pressure when washing the plate, as it can easily wash off the coating. Control the color development time; CV in low-value wells is easily affected by uneven temperature.

If delivery is required this week, contact the manufacturer's technical support or regional sales first, rather than just placing an order on the platform. Ask three things: whether it is in stock, whether the same lot number can cover 80 samples, and whether expedited cold-chain logistics can be provided. You can request an electronic version of the manual first to confirm that the sample types listed are serum/plasma and that a recommended dilution protocol is provided. If a certain brand is uncertain due to logistics, have two local distributors as backups. Within budget, it is not advisable to choose a 48T kit and split it for testing just to save money, because you need to run pilot experiments and replicate wells; a 96T kit gives you more flexibility.

If human serum IL-6 is being measured in clinical samples, healthy/non-inflammatory samples may have only a few pg/mL or even lower, and direct measurement with an ordinary kit can easily fall in the lower range of the standard curve. A reliable approach is not to fix the dilution factor in advance; instead, take 2-3 representative serum samples and test three conditions: neat, 2-fold, and 5-fold dilution, to see which falls into the middle range of the standard curve. If the sample volume is limited, a 2-fold dilution is preferable, as it both reduces matrix interference and leaves readable room for low values. For sensitivity, choose a lot with ≤1 pg/mL and pay attention to the lowest point of the standard curve, rather than only looking at the "limit of detection." The instructions saying direct measurement is possible does not mean all serum samples can be measured directly.

This week's arrivals need to be within 1500 per box. Prioritize finding in-stock distributors for major domestic manufacturers. Lianke, Sizhengbo, Xinbosheng, and Dayou have been making human IL-6 high-sensitivity/basic versions for a relatively long time, and distributors often have cold-storage stock, but have the sales rep send you the COA, standard curve range, and expiration date for that lot for confirmation first. Within 1500, imported high-sensitivity kits are basically out of the question, and imported standard versions may also exceed that. If there is a local bioreagent distributor, ask clearly whether cold chain is included and whether next-day delivery is possible. For 80 samples, it is recommended to buy the same lot of 96T at one time to avoid lot-to-lot variation.
