普通PCR试剂盒和荧光定量试剂盒不建议直接互换。qPCR除了模板和引物，还需要Mix里具备热启动酶、荧光染料或探针兼容体系、缓冲液离子强度与ROX/被动参考适配仪器。土壤样腐殖酸会抑制酶和荧光，优先选环境样品或抗抑制剂qPCR Mix，做标准曲线、熔解曲线、无模板对照和抑制内参；同一批样先小范围验证扩增效率和重复性再扩到300多个样。货期紧张可让供应商先寄试用装，按单次反应消耗核算，避免囤大包装临效期。

预处理比换Mix更关键。土壤DNA提取建议用商业化土壤或环境微生物试剂盒，必要时加物理珠磨和抑制剂去除步骤；洗脱后可关注A260/230和A260/280趋势，若腐殖酸残留明显，可稀释模板、加BSA或qPCR兼容抑制剂去除剂，但BSA会影响定量，需固定浓度并留对照。qPCR板设抑制对照：已知阳性模板加不加土壤DNA，观察ΔCt。300多个样建议先做9到12个梯度样，确定模板稀释度和Mix窗口，再批量跑板，降低返工风险。
