Primary neurons are more fastidious than tumor cells, so a phased release approach is recommended. First, have each of the three vendors provide trial samples from the same lot, and design a scoring table: lead time, appearance/precipitation, endotoxin, hemoglobin, neuronal support capacity (morphology and neurite length at 24/48 h after plating), and tumor cell proliferation. After arrival, perform rapid screening first: endotoxin/hemoglobin/sterility; once passed, proceed to primary validation: use the same batch of primary neurons and 2 types of tumor cells, each passaged 3 times, and compare attachment rate, doubling time, and functional indicators. Give suppliers clear terms for "lot locking" and "lot change validation." The purchase price can be negotiated first based on estimated annual volume, but place only a small initial order. Retain all validation samples in cryopreservation so that if differences arise later, the same lot can be traced.

Price comparison should not be based solely on the per-bottle quote; a gap of 2,000 to 6,000 often corresponds to origin, blood collection method, filtration, and batch-to-batch consistency. I suggest first asking three suppliers to provide a CoA and a 50 ml trial sample, with endotoxin, hemoglobin, total protein, origin, and whether it is inactivated clearly stated. When comparing prices, include validation costs: whether the trial sample is paid or free, minimum order quantity, cold-chain shipping, and whether batch changes are allowed. Before placing a formal order, first buy 2-3 bottles of the smallest packaging as retention samples, and freeze at least 2 bottles from the same batch, noting the lot number and arrival date. Specify in the contract that: for out-of-stock batch changes, a trial of the new batch must be provided in advance and validated; nonconforming batches can be returned or exchanged. In this way, low-priced batches that fail validation after testing will not be left on your hands.

Pre-purchase validation should be conducted at three levels. First, obtain 1-2 trial samples and culture them in parallel with the currently used imported serum on the same batch of cells. The first level involves physicochemical testing: endotoxin (LAL method), hemoglobin, sterility, mycoplasma, and precipitation/appearance; the second level involves cell proliferation: for primary neurons, use CCK-8/trypan blue to generate a 3-5 day curve, assessing neurite length and soma refractility; for tumor cells, assess doubling and colony formation; the third level involves functional testing: for primary neurons, stain MAP2/Tuj1 to assess neuronal proportion; for tumor cells, perform drug sensitivity or passage stability testing. Each brand should be tested independently at least twice, with the old serum as control. Endotoxin and hemoglobin are recommended but not the sole indicators; primary neurons are more sensitive to batch-to-batch component variation.

Cell growth curve, colony formation rate, mycoplasma and endotoxin testing—these three are mandatory.
