FAQ

Q：标准曲线扩增效率小于 90%或者大于 110%，线性关系不好，这样的问题如何解决？

A：加样误差。加大模板稀释倍数，提高加样体积，使不同稀释度的加样体积更准确；

标准品降解。重新制备标准品；

模板浓度太高。存在反应抑制，增加模板稀释倍数；引物扩增特异性不好。重新设计引物。

Q：反应结束无扩增曲线出现

A：反应循环数不够，可适当增加循环次数。一般设置为 40，需注意过多的循环会增加背景信号，降低数据可信度；

确认程序中是否设置了信号采集步骤。三步法扩增程序应该将信号采集设置在 72℃延伸阶段，两步法一般在退火延伸阶段收集；

确认引物是否降解。长时间未使用的引物应先通过PAGE 电泳检测完整性；模板浓度太低。减少稀释度重复实验；

模板降解。重新制备模板。

Q：实验重复性差如何解决？

A：加样体积不准。定期校正移液器，将模板高倍稀释，以大体积加入反应体系中；定量PCR 仪器不同位置温度控制不一致，定期校准仪器；

模板浓度太低。模板浓度越低，重复性越差；做 4-6 个复孔，删除其中重复性不好的孔。

Q：模板的稀释液如何选择？

A：Nuclease-Free water、DEPC 水或实验室自备的 ddH2O 都可以。不推荐使用 TE 作为稀释液，TE 里有EDTA 会抑制酶的活性。
