Without a Luminex instrument, CBA is a more realistic choice, because CBA only requires a flow cytometer to perform. Both BD CBA and BioLegend's LegendPlex offer mouse multiplex kits, with sample loading volumes typically 25-50 μL/tube, so measuring 4 targets from a few dozen microliters of serum can be arranged. The procedure takes about half a day: staining, washing, and acquisition. Sensitivity is not necessarily inferior to Luminex, especially since inflammatory factors are often not low; the key is the standard curve and sample dilution factor. It is recommended to first perform a dilution pilot test with a small amount of serum, such as 2-fold and 5-fold dilutions, to see which dilution falls within the linear range.

Luminex is more suitable for cases with multiple indicators, high throughput, or the need for a wider dynamic range, but both the instrument and kits are expensive. Without a machine, samples can only be sent out for testing, and the per-sample cost of send-out testing is also not low. CBA kits are relatively cheaper, and when a flow cytometer is already available, there is almost no additional equipment investment. In terms of sensitivity, Luminex usually has an advantage for low-abundance factors, but in mouse inflammation models, CBA is often sufficient for IL-2, IL-6, IFN-γ, and TNF-α. For operation time, both require preparing standards and plate washing; with CBA, the flow cytometry run time will increase with the number of samples. If the budget is limited, choose CBA first.

Also, be careful not to repeatedly freeze and thaw serum samples. For samples of a few dozen microliters, it is best to aliquot them into single-assay volumes immediately after collection and proceed directly to the assay workflow. For multiplex assays, the sample diluent and standard diluent must be matched; protein-containing buffers are commonly used to reduce matrix interference. When purchasing, ask the supplier to provide recovery/parallelism data for these factors in mouse serum. If you do not have a flow cytometer or do not want to perform the assay yourself, you can send samples to a commercial Luminex testing service, but you should likewise require them to report dilution recovery. When comparing options, factor in instrument occupancy, kit shelf life, and the number of replicate sample tubes loaded.

When the sample size is limited, Luminex is still the more suitable option, as CBA has relatively obvious limitations in terms of the amount required and the number of indicators.

We used to measure 5 analytes with CBA, and the serum was not enough so dilution was required, so we switched to Luminex.
