If it is only at the research stage, domestic products are indeed worth trying. A lead time within one week is achievable; manufacturers in East China are basically 3-5 days. However, pay attention to the batch-to-batch stability of domestic magnetic beads, especially activation titer and antibody density. It is recommended to order at least 2-3 batches for the first time, and retain samples from each batch for accelerated stability testing to avoid pre-clinical data jumping between batches. When purchasing, we simultaneously require: antibody clone number, binding capacity report, endotoxin, mycoplasma, and magnetic bead residue detection method. In terms of price, there are large differences among different specifications; the unit price difference between 1 mL and 5 mL packs is obvious. It is recommended to request quotes based on subsequent scale-up batch sizes, and not to estimate only based on the unit price of small samples. If, after domestic substitution, you plan to file an IND in the future, the raw materials must be able to support supplier audits and master file registration.

We have conducted a domestic substitution evaluation. The key point is not to look only at expansion fold. We recommend dividing PBMCs from the same donor into three groups: Dynabeads and domestic candidates A/B, with a bead-to-T cell ratio of 1:1 or 3:1. After activation, count on D3/D5/D7/D9, and simultaneously analyze CD4/CD8, CD45RA/CD62L, and PD-1/LAG-3/TIM-3 by flow cytometry. We encountered one domestic batch-to-batch difference that was mainly reflected in viability after D7 and the CD8 proportion, so at least two batches should be tested. Domestic beads have a small particle size and less residue after dissociation, and the impact on lentiviral transduction is generally manageable. For head-to-head experiments, do not look only at D9 expansion; it is recommended to extend to D12, because some domestic products expand quickly early and then drop later. In terms of cost, domestic products are about 30%-50% cheaper than Dynabeads; inquire specifically based on batch size.

Whether domestic CD3/CD28 magnetic beads can serve as substitutes hinges on four key points: antibody clone number and conjugation method, particle size, dissociation method, and endotoxin/magnetic bead residue. We have screened two vendors in Jiangsu and Beijing, with particle sizes ranging from 1.5 µm to 4.5 µm; smaller particle sizes are more favorable for subsequent magnetic bead removal. For dissociation, mild enzymatic digestion or degradable options are preferable; avoid strong mechanical scraping, as this disrupts T cell activation status and reduces transduction efficiency. Pay attention to endotoxin, mycoplasma, and sterility in the COA. Lead time is typically 3-5 working days for in-stock items, and the price for a 1 mL specification is, as I recall, in the range of several thousand RMB, considerably lower than imported products, but you must have the manufacturer provide a same-lot small sample for parallel testing with 3 donors. When selecting, ask clearly whether it can be used for clinical process development and whether there is CSV/regulatory support.

At the time, our biggest concern was that residual domestic magnetic beads would affect CAR lentiviral transduction. We recommend removing the beads before transduction. Regardless of whether they are imported or domestic, the residual bead amount should preferably be kept below the level recommended in the instructions. Many CD3/CD28 beads adsorb lentivirus, leading to inaccurate MOI. If domestic beads can be dissociated by biotin competition or enzymatic cleavage, the residual amount will be lower. Residual beads can be assessed by iron content testing or microscopic observation, and transduction positivity rates can be compared. We tested a batch of domestic alternatives. After bead removal on the day of transduction, the difference in CAR positivity rate compared with Dynabeads was within 5%, but in the same group, some colleagues did not remove the beads, and the positivity rate decreased by about 15%. Therefore, process integration is more critical than brand differences. We recommend small-scale validation: activate for 48-72 hours, remove magnetic beads, allow to stand for recovery, and then transduce.
