For stability, the key focus is freeze-thaw: some domestic master mixes claim to withstand 10 repeated freeze-thaw cycles, but in practice, with low-copy targets, multiple freeze-thaw cycles can affect Ct. After receiving a bulk shipment, it is recommended to aliquot into small tubes, with each tube sufficient for 1–2 experiments, to reduce repeated freezing and thawing of the entire bottle. When comparing prices, clarify whether the concentration of the 5 mL large pack is 2× or 5×; the per-well cost of 5× may be lower, but the sample loading volume must be adjusted. Also check whether ROX is included in the master mix or provided separately as a supplement, to avoid purchasing the wrong high/low version.

The ABI 7500 with high ROX is the conventional choice, but some domestic universal reagents may recommend low ROX or may work without ROX, so it is advisable to first confirm whether the instrument model is 7500 or 7500 Fast, and then check the compatibility table in the reagent instructions. Before bulk purchasing, it is best to have two domestic brands each send 5 ml for trial, and compare the Ct and amplification efficiency of the same batch of samples; if the budget allows, a small-pack imported reagent can be used as a reference. In terms of lead time, domestic products are basically within one week, imported products take 2-4 weeks, and tender procurement requires additional time.

For your 200 reactions per run, a 5 ml master mix is enough for about 500 wells when using a 20 μl system in a 96-well plate. After accounting for loss, that may cover only 2-3 experiments, so ordering 6-10 ml is safer. When comparing domestic and imported products, calculate cost per reaction, but imports may have slow delivery and cold-chain issues. For several mainstream domestic brands, you can first request 5 ml trial samples, run standard curves and melting curves on the same batch of cDNA, and calculate Ct SD. At the same time, ask clearly whether the 5 ml bulk pack is from the same lot and whether a CoA and retention samples can be provided.

In terms of discount channels, for universities and research institutes, purchasing through platforms or regional agents is more reliable than buying in small quantities, and you can negotiate payment terms and tiered pricing. A volume of 200 reactions is not particularly large, but if it accumulates to several thousand reactions in the second half of the year, you can ask the manufacturer for an annual agreement. Don't be attracted by "cheap premix"; check whether it is compatible with high ROX, whether it contains ROX, and whether you need to provide it yourself. I usually set aside a 2-week validation period and ask the supplier to provide recent batch performance data and a return/exchange commitment.

For the ABI 7500, it is recommended to choose the high ROX version; low ROX may result in insufficient baseline correction, affecting Ct and ΔRn. However, kits labeled as "universal ROX" by different brands also vary. The first thing to do after purchase is to run a standard curve with the same standard to check whether the amplification curve baseline is flat. For bulk procurement, do not focus only on unit price; some domestic 5 ml large-pack products can arrive within a week, while imported products may have long lead times. When comparing, the delivery cycle, inter-batch CV, freeze-thaw stability, whether ROX is included, and whether the instructions state 7500 compatibility should all be taken into account.
