The budget should not exceed 1500 yuan per vial. It is recommended to first purchase small packaging or trial sizes and perform parallel comparison in the same batch of sections with the generic type currently in use. Using the same tissue and the same retrieval conditions, incubate the two secondary antibodies separately to assess nonspecific and positive intensity. If the goal is to reduce nonspecific staining, prioritize pre-adsorbed or F(ab')2 fragment secondary antibodies; the price will be slightly higher but it is worth it. The consumption for 60 sections is not large, so small packaging is sufficient; do not stock up on large packaging all at once to avoid batch-to-batch variation.

Don't assume it's a secondary antibody problem by default. For paraffin section immunohistochemistry, strong non-specific staining is often more closely related to endogenous enzyme blocking, antigen retrieval methods, and blocking agents. For endogenous HRP blocking with 3% H2O2, do not treat for too long; antigen retrieval should be sufficient but not excessive, and the type of retrieval solution should be adjusted according to the target. For blocking, use 5% normal serum from the same species as the secondary antibody for 30 min at room temperature. Run a parallel control with no primary antibody and only secondary antibody to determine whether the background comes from the secondary antibody itself.

If you are only doing single-labeling, separate anti-rabbit and anti-mouse HRP secondary antibodies usually give more controllable background than a universal type, because universal types may cross-react with non-target IgG in order to accommodate multiple species. Dual-labeling kits are suitable for experiments that clearly require simultaneous rabbit and mouse double staining on the same slide; for routine work with only one target, there is no need to use a dual-labeling kit, as it is costly and the dilution conditions are inflexible. It is recommended to first switch to single-species secondary antibodies, perform a gradient of titers, and one small pack is sufficient for 60 sections.

I use both domestic and imported ones. For domestic HRP secondary antibodies such as Proteintech, Abclonal, and Beyotime, concentrated formats mostly cost several hundred yuan, and you can get a good one within a budget of 1,500. Imported brands like Vector, Jackson, and CST are often close to or above 1,500, but distributor discounts can sometimes bring them within budget. In terms of performance, it is not accurate to simply say imported ones are always better; domestic products have improved markedly in batch-to-batch consistency in recent years. The key is whether there are validation data for the corresponding paraffin sections and low-background batches.

Batch-to-batch consistency matters more than a low price alone. Ask suppliers for batch-specific test reports or inter-batch consistency documentation, and inquire whether other labs have feedback using the same batch for IHC. Imported products may take 3–7 days to two weeks to arrive, so stock up in advance. You can ask the distributor to reserve same-batch inventory, or buy two vials from the same batch at once. Upon receipt, check that the vial body has no leakage and the label is clear, and avoid repeated freeze-thaw cycles. Within this budget, you can choose imported or domestic secondary antibodies with consistent low background, but you must validate them yourself.
