Color panel design is not just about whether fluorescence channels conflict; compensation and gating must also be considered. It is recommended to prepare at least three single-stain compensation tubes for each batch of samples (CD3, CD4, and CD8 fluorescence separately). If using BV or APC-Cy7 tandem dyes, it is best to also prepare FMO tubes to confirm the gating boundaries for CD4 and CD8. In splenocytes, CD4 and CD8 populations are relatively distinct, but weak CD4-positive monocyte/DC background can interfere, and FMO can help exclude this. For tandem dyes, be careful not to fix overnight, as this may cause uncoupling and lead to false-positive spillover in APC-Cy7.

When preparing the spleen, first grind it with a 70 μm mesh, lyse the red blood cells, and then count. The viability should ideally be >90%; a high number of dead cells will significantly increase nonspecific staining and background. Staining with 100 μL per tube for 1×10^6 cells is fine, but be sure to perform Fc blocking first, then add the antibody cocktail, and incubate at 4°C in the dark for 30 min. The antibody cocktail can be premixed based on the total sample plus 10% loss to avoid pipetting errors between tubes. After staining, wash once with PBS containing 2% FBS, and finally resuspend for acquisition. If acquisition cannot be done the same day, fixation with 1% paraformaldehyde at 4°C in the dark is acceptable, but do not exceed 24 hours, and for tandem dyes, it is recommended to acquire as freshly as possible.

For mouse spleen phenotyping of CD3/CD4/CD8, if the instrument only has 488 and 640 lasers, a classic combination can use CD3-PerCP-Cy5.5, CD4-FITC, and CD8-PE. Compensation is mainly between FITC and PE, and it is still relatively easy to adjust when splenocytes show clear population separation; however, if testing weak expression or adding IL-17, etc., it is recommended to use BV421/BV510 with the 405 laser as much as possible. For example, CD3-BV421, CD4-FITC, CD8-APC, or CD3-PerCP-Cy5.5, CD4-APC, CD8-PE-Cy7. For each tube, 1×10^6 cells in a 100 μL system is sufficient. The key is to titrate the antibodies first according to the test amount, and do not blindly add more.

The manufacturer-recommended test amount is typically designed for 1×10^6 cells/100 μL, commonly 0.25 μg or 5 μL of stock solution per test, but this varies across brands and fluorophores. It is recommended to perform a 3-4 point titration after receiving the antibody: start with 1×, 0.5×, 0.25×, and 0.125× of the manufacturer-recommended amount, and evaluate the staining index of CD3+, CD4+, and CD8+ populations. Before staining splenocytes, always block with CD16/32, incubate at 4°C in the dark for 30 min, and wash once; do not use less than 50 μL in a 100 μL system, as overly concentrated cells will increase nonspecific binding. If background is high, add 2% FBS or BSA.

Domestic BD, BioLegend, and Invitrogen/eBioscience regular clones such as 17A2, RM4-5, and 53-6.7 are often in stock, but special conjugates like BV421 and APC-Cy7 may require ordering. Generally, in-stock items take 3-7 days, while orders can take anywhere from 2-6 weeks. Before placing an order, have the distributor confirm domestic warehouse inventory and expiration dates; do not rely solely on the catalog. For small-package trial sizes, proactively ask the regional sales representative. 25 μg or test-size formats are typically used for validation, and some brands offer trials, but this requires lab information and a description of the intended use, and not all clones are available. If urgent, you can first borrow aliquots from other labs for preliminary experiments.
