It is somewhat reasonable for suppliers to say that the COA is only for release reference, because the COA usually tests only a limited set of indicators, and the sample may not represent the entire batch. To assess batch-to-batch stability, you should not rely only on total protein. You can ask the supplier for trend charts of historical batches, or accumulate data from 3-5 consecutive batches yourself, calculate the mean ± SD, and set internal control alert limits. If total protein fluctuates beyond the historical range or affects key indicators, such as a decrease in transfection efficiency of more than 20%, then return the product or switch to another batch. At the same time, pay attention to endotoxin, hemoglobin, pH, etc., and establish your own batch release standards.

Total protein ranging from 4.2 to 5.8 g/dL, a difference exceeding 30%, is relatively large for the same brand. 293T transfection is more sensitive to lipids, complement, growth factors, etc. in serum. Higher total protein may be accompanied by more unknown proteins, which may affect the stability of liposome or PEI transfection complexes. It is recommended not to use it directly for critical experiments. First perform small-scale validation: revive a small amount from each of two lots, culture 293T in parallel, and compare cell doubling, transfection efficiency, and cell viability. If the difference is within an acceptable range, then release it.

For one purchase of 20 bottles, it is recommended not to thaw all of them. First keep 2-4 bottles for small-scale validation, and keep the rest at -20°C or -80°C. In addition to routine sterility and mycoplasma testing, the validation items should focus on 293T transfection efficiency, preferably using the transfection reagent and plasmid commonly used in your laboratory. At the same time, retain at least 1-2 bottles from each batch as long-term retention samples, so that if abnormal data appear later, traceability can be carried out. When receiving into inventory, record the lot number, arrival date, and COA number, and require the supplier to provide the original factory inspection report or a stamped scanned copy.
